The test is based on the fact that udder irritation or inflammation caused by mastitis results in an increased concentration of white blood cells in the milk. These cells form a gel when mixed with Mastest – the degree of gelling being closely related to the concentration of cells and, with a few exceptions, to the degree of irritation or inflammation.
